چکیده

Intracranial hypertension (IH) is a rare condition in children. However, a relationship between recombinant human growth hormone (rhGH) therapy and IH has been well documented. Risk factors were assessed for 70 rhGH-naive patients enrolled in the National Cooperative Growth Study with reports of IH after treatment initiation. Patients with severe growth hormone deficiency, Turner syndrome, chronic renal insufficiency (CRI), and obesity (particularly in the CRI group) were at highest risk of developing IH during the first year of therapy, suggesting initiation of careful early monitoring. In some patients, factors such as corticosteroid use or other chromosomal abnormalities appear to confer a delayed risk of IH, and these patients should be monitored long-term for signs and symptoms of IH.

Lessons from the national cooperative growth study.

OBJECTIVE To review the National Cooperative Growth Study (NCGS), a national post-marketing surveillance program for children treated with biosynthetic growth hormone (GH) products from Genentech. METHODS Representative data are presented to summarize the extensive experience of the NCGS. This study is a multicenter, observational surveillance registry begun in 1985 in coordination with the r...

Therapeutic Effects of Recombinant Growth Hormone in Improving the Growth of Thalassemia Major Patients: A Retrospective Study

Background and purpose: Beta thalassemia major is one of the most common inherited blood diseases and patients with this condition require regular blood transfusions. The resulting iron overload deposition causes endocrine gland disorders, including growth hormone (GH) abnormality. In this study, we examined the effects of recombinant growth hormone (rGH) in these patients. Materials and metho...

Papillary Carcinoma of Thyroid Gland in a Patient Treated with Recombinant Growth Hormone

The first successful treatment of growth hormone (GH) deficient children with GH extracted from human pituitary was introduced during late 1950's.  The subsequent availability and use of recombinant GH (rhGH) for different clinical conditions raised the question of whether this new therapeutic modality increases the risk of certain conditions such as leukemia or malignancy.  Herein, we report o...

Comparison of Growth Impairment in Pediatric Patients with ALL, NHL Treated with Chemotherapy alone and Chemoradiotherapy

Introduction: Acute Lymphoblastic Leukemia (ALL) and Non Hodgkin’s Lymphoma(NHL) are the most common cancers in childhood and  adolescence. Most children with ALL and NHL have long term survival (>80%). Endocrinopathy is a common complication in pediatric cancers following the treatment (20-50% prevalence), in addition, cytotoxic chemotherapy and radiotherapy can influence side effect on growt...

CONSTRUCTION OF RECOMBINANT PLASMIDS FOR PERIPLASMIC EXPRESSION OF HUMAN GROWTH HORMONE IN ESCHERICHIA COLI UNDER T7 AND LAC PROMOTERS

In order to study the periplasmic expression of human growth hormone (hGH) in Escherichia coli, the related cDNA was inserted in two expression plasmids carrying pelB signal peptide, one with lac bacterial promoter and the other with a bacteriophage T7-based promoter. The recombinant plasmids were moved to TG1 and BL21 strains of E. coli, respectively. To induce the expression systems, IPTG and...
